About the Position
We are looking for a PhD level scientist that can thrive in a fast-paced, dynamic organization with the goal of therapeutic antibody discovery and development. The selected candidate will work in the Protein Science group in close collaboration with a team of biologists and engineers based at HiFiBiO Cambridge, Paris and Shanghai sites. The candidate will be responsible for leading the efforts on antibody engineering, reformatting, humanization, and affinity maturation. The candidate will help in designing and engineering different mono-specific, bispecific, and fusion antibody molecules that can be evaluated for various HiFiBIO programs In addition to antibody engineering, the candidate will design and perform experiments to characterize antibodies, including epitope binning and mapping, affinity measurement, sequence liability identification, immunogenicity prediction, therapeutic developability assessment using Octet, chromatography, ELISA and other biochemical and bioanalytical methods.
The responsibilities include but are not limited to:
- Engineering, expression and purification of monoclonal mono- and multi-specific antibodies and fusion proteins.
- Design and execution of humanization and affinity maturation strategies.
- Documenting experiments, results and findings in electronic laboratory notebook.
- Teaching and mentorship of junior research associates and interns, working with collaborators and CROs.
- Expertise in molecular, biochemical, biophysical and bioanalytical techiniques such as PCR amplification, mutagenesis, molecular cloning and plasmid design, SEC, light scattering, SDS-PAGE, western blot, ELISA and BLI (Octet).
The ideal candidate holds a PhD degree in Biophysical Chemistry, Molecular Biophysics, or Molecular Biology / Biochemistry. Post-doctoral training of 1-3 years is preferred but not required. The candidate is expected to have proven expertise in protein or enzyme engineering, demonstrating strong problem-solving ability via biochemistry, structural biology, protein engineering or computational biology methods. Experience characterizing and engineering proteins or enzymes using analytical biochemical and biophysical methods is preferred. Structure based protein design experience is highly valued. Experience with any one of the display platforms (phage/Yeast/mRNA), is highly valued. Strong publication record in related fields is required.
